The second most produced fruit on a worldwide scale of tropical fruits is citrus. Crucial dietary micronutrients are found in abundance in citrus fruits. Consuming sufficient citrus fruits and their juices, which are rich in vitamin C, phytochemicals, and antioxidants improves health, lowers the risk of non-communicable diseases, provides the best nutrition, and promotes overall wellbeing. Antioxidant molecules found in fruits with excellent antioxidant qualities can reduce the negative effects of free radicals on our bodies. The quality attribute parameters such as juice content (43.50 to 50.84%) and (37.89 to 48.02%), pH (3.72 to 3.92) and (2.52 to 2.62), titratable acidity (0.64 to 1.24%) and (1.62 to 1.96%), total soluble solids (9.29 to 9.73 °Bx) and (7.79 to 8.47 °Bx), and TSS: TA ratio (7.48 to 15.19) and (4.32 to 4.80) was obtained for oranges and lemons respectively. The antioxidant potential was determined by measuring vitamin C, total phenolic content, total flavonoid content, and 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity using a spectrophotometer. The vitamin C ranged from 2.87 to 6.21 mg/mL for oranges and 9.89 to 12.49 mg/mL for lemons. The TPC ranged from 27.34 to 46.10 mg GAE/L for oranges and 24.85 to 26.99 mg GAE/L for lemons. The TFC of orange extracts was 0.4379 to 0.4581 mg QE/L extract and for lemons, it was 0.4336 to 0.4564 mg QE/L extract. DDPH radical inhibition activity ranged from 12.93 to 93.57% for Awash orange and 14.33 to 95.65% for Bahir Dar orange, and for lemons, from 19.16 to 95.46% for Awash lemon and 9.86 to 73.66% for Bahir Dar lemon. Hence, Bahir Dar orange and Awash lemon extract have more potent free radical inhibiting activity. The extract's antioxidant activity was also evaluated using the IC50 method. Linear regression was used to determine it. The IC50 values for Awash and Bahir Dar oranges were 4.66 and 3.93 mg/ml, respectively, while they were 4.09 and 6.44 mg/ml for Awash and Bahir Dar lemons. The more powerful the antioxidant activity, the lower the IC50 value. Awash lemon and Bahir Dar orange extract have more potential for antioxidant activities.
